Integrin α IIb β 3 Activation and Clustering in Minimal Synthetic Cells
نویسندگان
چکیده
Platelet adhesion and activation are mediated by integrin αIIbβ3 clustering, which is crucial for the hemostatic function of platelets. In an activated state, integrins provide connection between extracellular matrix actin cytoskeleton through a variety cytoplasmic proteins, such as talin. Here, droplet-based microfluidics applied to generate cell-sized giant unilamellar vesicles (GUVs) with defined molecular composition quantify αIIbβ3-containing protocells in relation number integrin–talin head domain (THD) complexes. Furthermore, it shown that THD induces clustering adhering fibrinogen. The formation this link, has, so far, only been observed vivo, essential step synthetic cell design recapitulate integrin-mediated bidirectional signaling across membrane. These results pave way further quantitative investigations protein–protein interactions associated proteins their assembly within defined, but complex, cells. An future mimic complex interaction cells environment will be combine approaches peptide chemistry guide mechanisms involved binding activation.

Minimal Synthetic Cells to Study Integrin-Mediated Adhesion
To shed light on cell-adhesion-related molecular pathways, synthetic cells offer the unique advantage of a well-controlled model system with reduced molecular complexity. Herein, we show that liposomes with the reconstituted platelet integrin αIIb β3 as the adhesion-mediating transmembrane protein are a functional minimal cell model for studying cellular adhesion mechanisms in a defined environ...
متن کاملDisruption of disulfide restriction at integrin knees induces activation and ligand-independent signaling of α₄β₇.
Control of integrin activation and signaling plays crucial roles in cell adhesion, spreading and migration. Here, we report that selective breakage of two conserved disulfide bonds located at the knees of integrin α4C589-C594 and β7C494-C526 activated α4β7. This activated integrin had a unique structure that was different from the typical extended conformation of active integrin. In addition, t...
متن کاملIntegrin α 3 β 1 directs the stabilization of a polarized lamellipodium in epithelial cells through activation of Rac 1
Cell migration is a crucial event that occurs during many processes including wound healing, angiogenesis, development and metastasis. Although migratory phenotypes and factors that promote migration vary greatly among cell types, the basic mechanism that drives cell migration in all cells is polarized actin polymerization (Pollard and Borisy, 2003). This process is initiated by chemotactic fac...
متن کاملIntegrin IIb 3 Activation in Chinese Hamster Ovary Cells and Platelets Increases Clustering Rather than Affinity*
Integrin IIb 3 affinity regulation by talin binding to the cytoplasmic tail of 3 is a generally acceptedmodel for explaining activation of this integrin inChinese hamster ovary cells and human platelets. Most of the evidence for this model comes from the use of multivalent ligands. This raises the possibility that the activationbeingmeasured is that of increased clustering of the integrin rathe...
